DNA Polymerase I from E. coli is a DNA-dependent DNA polymerase. The enzyme also contains both 5´→3´ and 3´→5´ exonuclease activities.3 The 5´→3´ exonuclease activity enables the enzyme to use nicks and gaps in the DNA as starting points for labeling DNA by nick translation.
Unit Definition: One unit of DNA Polymerase I, E. coli, converts 10 nmol of dNTPs into acid-insoluble material in 30 minutes at 37°C under standard assay conditions.
Storage Buffer: 50% glycerol containing 50 mM Tris-HCl (pH 7.5), 0.1 M NaCl, 0.1 mM EDTA, 1 mM DTT, and 0.1% Triton® X-100.
Quality Control: DNA Polymerase I, E. coli, is free of detectable contaminating endonuclease and RNase activities.
