Ribonuclease T1 (RNase T1) is an endoribonuclease that specifically cleaves RNA or deaminated RNA at the 3´ end of guanosine residues and adjacent nucleotides through a 2´,3´ cyclic phosphate intermediate mechanism. Epicentre’s RNase T1 is cloned from Aspergillus oryzae and overexpressed in E. coli to produce a highly pure enzyme without contaminating DNase or nonspecific RNase activity.
Unit Definition: One unit of RNase T1 converts 100 ng of E. coli ribosomal RNA into acid-soluble nucleotides in 5 seconds at 37°C under standard assay conditions.
Storage Buffer: 50% glycerol containing 50 mM Tris-HCl (pH 7.5), 100 mM NaCl, and 0.1 mM EDTA.
Quality Control: RNase T1 is free of detectable DNA exo- and endonuclease activities and non-RNase T1 RNase activity.
